Autoclave Loading and Cycle Selection: Liquids, Solids and Waste

Autoclave loading decides whether a cycle works, and the load decides which cycle to run. At LabSupplies.com, most of the autoclave questions that reach us are not about which machine to buy — they are about a cycle that ran, logged a normal record, and still left a bagged waste load warm in the middle or a flask of media boiled across the chamber floor. Both failures come from the same place: the chamber hit temperature and the load did not, or the load was depressurised faster than it could cool.

Four Load Types, Four Different Cycles

That matters most in a shared lab, where the person running the machine at 6 p.m. is not the person who wrote the method.
Liquids: the Failure Everyone Has Seen
A liquid at sterilisation temperature sits above its atmospheric boiling point. Release chamber pressure quickly and it flashes to steam inside the bottle. That is why the liquid course exhausts slowly, and why running media on an instrument course produces the classic mess of boiled-over agar and half-empty flasks.
The loading rules follow from the same physics:
- Fill vessels to no more than about two thirds, so there is headroom for expansion and bubbling.
- Loosen or vent every closure. A sealed bottle is a pressure vessel inside a pressure vessel.
- Scale the cycle to the largest single volume, not the total. A 2 L flask needs materially longer at temperature than four 500 mL bottles.
- Stand vessels upright in a container that will hold the contents if one does fail.
That last point is what the solid stainless buckets are for. A mesh basket is the right choice when steam has to reach the load from every direction; a solid bucket is the right choice when something might boil over or drip.
Baskets, Buckets and Plates
Load-carrying hardware is model-specific on these units, and buying the wrong size is a common and avoidable error — the accessories are listed against the exact chamber families they fit.

Whatever you load into, leave gaps. Packing a chamber tight is the single most common cause of a cycle that logs correctly and still fails in the middle, because steam has to displace air from every surface it needs to reach. Do not rest containers directly on each other, do not stand trays flat where they will hold a puddle of condensate, and do not fill the chamber to the roof because the basket allows it.
Monitor the Load, Not the Chamber
The chamber probe tells you the cycle ran. It does not tell you that the centre of a 2 L bottle or a dense bag of waste got there. On the Yamato SN and SQ families the difference can be instrumented directly: a sample temperature sensor is a Type T thermocouple placed in the load itself, and a chamber temperature sensor set supplies three probes for mapping the chamber.

The habit worth building is simple: use the load probe once for each new load configuration you intend to run routinely, record what the centre of that load actually did, then trust the chamber record for repeats of that same configuration. Change the container, the fill volume or the packing density and you have a new configuration.
Waste Loads Are Their Own Problem
Autoclaving regulated waste is treatment, not tidying, and the requirements around handling and labelling that waste sit in the OSHA bloodborne pathogens standard. Mechanically, the challenge is that a bag full of waste is a bag full of insulating air.
- Leave the bag open or vented — a sealed bag will not sterilise reliably, however long the cycle runs.
- Add a small volume of water to generate steam inside the load.
- Load in a mesh basket inside a containment tray, so a melted bag does not reach the chamber.
- Extend the cycle for density. Waste loads need longer than the same mass of loose glassware.
- Keep waste cycles and clean cycles as separate documented courses, not as one course someone adjusts.

Drying, and Why Wet Packs Matter
A wrapped pack that comes out damp has lost its barrier: moisture wicks contamination straight through the wrap. If your workflow needs loads ready to use, an integrated drying stage is worth the price difference — the Yamato SM series adds a 150–180 °C drying stage after sterilisation. Without a dryer, build in air-drying time and do not stack packs while they are still warm.
Where steam is the wrong medium entirely — powders, oils, sealed glassware — the answer is a dry-heat sterilizer rather than a longer steam cycle. Before You Open the Door
Yamato units interlock the lid so it cannot be opened under pressure, and add pressure relief, dry-run prevention and self-diagnostics. Those protect the machine and the operator from the obvious failure. They do not protect anyone from a bottle of superheated liquid that looks still and is not: let liquid loads stand before you move them, wear the right gloves and eye protection, and treat a chamber that has just finished a liquid course as hot rather than done. Frequently Asked Questions
Why does an autoclave need a separate liquid cycle?
Because of how the chamber is depressurised at the end. An instrument cycle can exhaust quickly; a liquid load cannot. Liquids at sterilisation temperature are above their atmospheric boiling point, so a fast pressure drop makes them flash-boil and erupt out of the vessel. A liquid course exhausts slowly enough for the load to cool below boiling before the pressure is released.
How full should bottles and flasks be in an autoclave?
No more than about two thirds, with closures loosened or vented. A tightly capped bottle is a pressure vessel inside a pressure vessel, and the headroom is what keeps a boiling liquid from carrying over. Larger volumes also need longer cycles, because the time is set by how long the centre of the liquid takes to reach temperature, not by the chamber.
Should I use a mesh basket or a solid bucket?
Mesh for anything that needs steam to reach it from all sides, such as wrapped instruments, glassware and bagged waste. A solid stainless bucket is for loads where you want to contain liquid, either because a vessel may boil over or because the material must not drip into the chamber. Adjustable perforated plates let you layer a mesh basket without stacking items directly on each other.
Is chamber temperature the same as load temperature?
No, and the difference is the whole point of monitoring. The chamber reaches setpoint long before the centre of a large liquid volume or a dense pack does. A sample temperature probe placed in the load reports what the load actually experienced, which is what determines whether the cycle worked.
Can I autoclave biohazard bags in a normal cycle?
Only if steam can get inside the bag. A sealed polypropylene bag traps air, and trapped air is an insulator that keeps the contents below sterilisation temperature. Leave the bag open or vented, load it in a mesh basket inside a containment tray, add a small volume of water to help steam penetration, and extend the cycle for the density of the load.
