Brightfield microscopy requires specimens to absorb light — staining, fixing, or otherwise altering the sample before observation. Living cells, unstained cultures, sperm motility assessment, and other transparent specimens produce almost no contrast under brightfield. Phase contrast microscopy converts the invisible phase shifts that transparent specimens impose on transmitted light into visible contrast differences — live, unstained samples become observable without chemical treatment or fixation.
The VEE GEE Scientific Vanguard 1300 PHI Series (1323PHI binocular; 1333PHI trinocular) provides three imaging modes in one instrument: phase contrast for transparent live specimens, darkfield for motility and particle visualization, and standard brightfield for stained specimens. Plan achromatic objectives (10X/20X/40X/100X), 20W variable quartz halogen with Koehler illumination capability, and the same 5-year warranty as the full Vanguard 1300 line.
Features and Benefits
Three Imaging Modes — Phase Contrast, Darkfield, Brightfield: Switch between modes by rotating the condenser turret — phase contrast for live cells, darkfield for motility and surface structure, brightfield for stained slides. One instrument, three contrast methods.
Phase Contrast for Live, Unstained Specimens: Observe living cells, bacteria, yeast, tissue culture, and aquatic organisms without fixation or staining — specimens remain viable and dynamic during observation.
Plan Achromatic Objectives (10X/20X/40X/100X): Flat-field plan correction across all four objectives. The 20X objective expands the magnification range compared to the standard 4X/10X/40X/100X brightfield configuration — useful for tissue culture and large-specimen observation.
Trinocular Option (1333PHI): Third optical port for VEE GEE 1300-CDPC digital cameras — enables live image capture and video documentation of phase contrast specimens without removing the camera to view through the eyepieces.
Koehler Illumination: 20W variable quartz halogen with field diaphragm — Koehler setup optimizes phase annulus alignment and condenser positioning for correct phase contrast operation.
5-Year Warranty: Full 5-year coverage on optics, phase condenser, and illumination system.

Laboratory Applications
Cell biology and tissue culture — live cell observation, mitosis staging, and morphological assessment of cultured cells without fixation
Microbiology — direct observation of bacterial motility, yeast budding, and protozoan movement in live wet mounts
Reproductive medicine and veterinary fertility — sperm motility analysis and morphology assessment using phase contrast for unstained semen samples
Teaching and research where both fixed/stained (brightfield) and live/unstained (phase contrast, darkfield) specimen observation are required in the same course or workflow
Frequently Asked Questions
What is the difference between 1300 PHI and 1300 BRi series?
The 1300 BRi (1321BRi/1331BRi) is brightfield only with 4X/10X/40X/100X objectives. The 1300 PHI (1323PHI/1333PHI) adds phase contrast and darkfield capabilities via a phase condenser turret, and uses 10X/20X/40X/100X objectives (replacing 4X with 20X for better tissue culture observation). Choose BRi for stained-specimen clinical and educational use; choose PHI for any application requiring live, unstained specimen observation.
Do I need to purchase phase objectives separately?
No — the 1323PHI and 1333PHI include the phase condenser and all phase-compatible objectives. The complete phase contrast configuration ships as a ready-to-use instrument.
Can the 1333PHI capture phase contrast video?
Yes — with a compatible VEE GEE 1300-CDPC USB camera attached to the trinocular port, both still image and video capture are available in phase contrast mode. The imaging software included with the camera supports live video preview and video recording.
Quality Assurance
VEE GEE Scientific Vanguard 1300 PHI Series microscopes are manufactured with factory-aligned phase condenser annuli and plan achromatic phase objectives. Phase annulus alignment is verified at the factory for each condenser position. 5-year warranty covers optics, phase condenser, and illumination system.
