A tube cap is the cheapest component in the specimen pathway and the one most likely to ruin it. Start With Diameter, Not Style

Closures are sold by the tube's outside diameter, and the common lab sizes are tightly clustered: 10, 12, 13, 16 and 17 mm. Measure the tube OD rather than inferring it from the nominal volume — a 5 mL and a 10 mL tube can share a diameter, and two 15 mL tubes from different lines may not.

Several formats are cut to span more than one size. The practical rule: if the tube will be inverted, shipped, or frozen, use a screw cap or a flange plug. A plain snap cap is a workflow convenience, not a transport seal.

Cold Storage Changes the Answer

Polymers stiffen and contract as they go down, and a closure that seals at bench temperature can loosen at −80 °C. Cryogenic caps are specified separately for that reason. When the Cap Has to Be Optically Clear

Most closures never need to transmit light. PCR is the exception. On a real-time instrument the optics read through the cap, so the closure is part of the optical path — a standard opaque cap makes the run unreadable.

Colour as a Working Control

Most of these closures are stocked in a colour range — blue, green, red, yellow, lavender, white, grey, orange, black and natural — and the colour is doing real work, not decoration. Colour-coding by assay, by draw time, by patient panel, or by study arm turns a rack of visually identical tubes into something a technician can sort at a glance and a supervisor can audit from across the bench.

The one caution: colour conventions are local. A scheme that is obvious to the person who set it up is invisible to a night-shift locum, so write it down and post it where the tubes are capped.

Pack Sizes and What They Imply

Closures are consumables bought deep — typically 1,000 per bag, with case quantities running to 10,000 or 20,000, and cryo caps in smaller 200-per-bag counts because the use rate is lower. Two things follow. First, a closure is not a trial purchase; confirm fit on a sample before committing to a case. Second, because the per-unit cost is low and the failure cost is high, this is the wrong line to economise on by substituting an approximate fit.

Frequently Asked Questions

How do I work out what size cap my tubes need?

Measure the tube's outside diameter at the rim, in millimetres. Closures are sold by that dimension — 10, 12, 13, 16 and 17 mm are the common lab sizes — and not by the tube's nominal volume, which is not a reliable guide to diameter.

What is the difference between a snap cap, a plug cap and a flange plug cap?

A snap cap grips over the outside of the tube rim. A plug cap pushes down inside the bore, giving a lower profile and a tighter seal against evaporation. A flange plug does both: the plug seals inside while a flange rests on the rim and gives you something to grip when removing it.

Are tube caps interchangeable between tube brands?

Only where the manufacturer states compatibility. Diameter alone is not sufficient, because rim geometry and wall thickness vary between lines. Some closures are explicitly specified as BD®-compatible; where no compatibility is stated, trial-fit before ordering a case.
