Carboys for the Lab: Selection, Spigots and Safe Handling

Carboys are chosen on four things: resin, mouth width, spigot or no spigot, and shape. Get the resin right first — it determines what you can store and whether you can autoclave. Polypropylene is autoclavable and handles a broad chemical range. HDPE and LDPE are not autoclavable but cost less and handle aqueous media well.

Resin selection

Polypropylene (PP)
The choice when the carboy must be autoclaved. PP holds shape through steam cycles, resists a wide range of acids, bases and alcohols, and is the standard for media preparation and buffer storage that has to be sterilized in the vessel. Round PP carboys and heavy-duty PP versions cover most of this work.
High-density polyethylene (HDPE)
Stiffer and more chemically resistant than LDPE, and the usual pick for bulk water, aqueous reagents and waste collection. Not autoclavable. Amber HDPE adds light protection for photosensitive solutions — worth specifying for anything that degrades under UV rather than wrapping a clear carboy in foil.
Low-density polyethylene (LDPE)
More flexible and translucent, which makes level-checking easier. Lowest cost of the three and fine for DI water and general aqueous use. Not autoclavable, and the flexibility that helps you see levels also means it deforms more readily under load when full.
Wide mouth or narrow mouth
Narrow mouth reduces evaporation and spill risk in transport, and gives a more secure seal. Wide mouth is the right call whenever the carboy has to be cleaned — you cannot get a brush or your hand into a narrow neck, and biofilm in a bulk water carboy is a real contamination route. If the vessel holds anything that leaves residue, go wide mouth.
Spigots: convenience against risk

A spigot carboy means you dispense without lifting, which is the single biggest ergonomic win at 10–20 L. The tradeoff is that a spigot is an additional seal, and it is the part that fails. Two rules that prevent most incidents:
- Never transport a carboy by the spigot or with the spigot loaded. Move it upright and empty where possible.
- Do not autoclave a carboy with the spigot fitted and closed. Pressure has to equalize, and a sealed spigot assembly is a failure point. Remove or loosen it per the manufacturer instructions.
Spigots are consumable. When one starts to weep, replace it rather than overtightening — replacement spigots are available separately.
Round or rectangular
Rectangular carboys use bench and shelf space far more efficiently and sit stably against a wall, which is why rectangular spigot carboys are popular for benchtop DI water. Round carboys are stronger for a given wall thickness, pour more cleanly, and are easier to clean because there are no corners to trap residue. Choose rectangular for stationary dispensing, round for anything that moves or needs thorough cleaning.

Handling and safety
A full 20 L carboy weighs over 20 kg before you count the vessel. That is squarely in the range where NIOSH lifting guidance says manual handling becomes a musculoskeletal injury risk, particularly with the awkward grip and shifting liquid load a carboy presents — see the NIOSH ergonomics and musculoskeletal disorders guidance. Practical mitigations: dispense via spigot rather than pouring, use a carboy cart or dolly for transport, and fill to a working volume you can actually handle rather than to the brim.

Waste carboys must stay closed except when adding waste, and must be labeled with contents and hazards — an unlabeled carboy under a fume hood is a common inspection finding.

Can carboys be autoclaved?
Polypropylene carboys can be. HDPE and LDPE cannot — they deform at steam sterilization temperature. Always remove or loosen the closure and spigot so pressure can equalize.
Should I get a spigot?
Yes for stationary bulk dispensing at 10 L and above — it removes the need to lift and pour. No if the carboy is transported frequently or autoclaved often, since the spigot is an extra seal and the usual failure point.
What does an amber carboy do?
Amber resin blocks UV and short-wavelength visible light, protecting photosensitive solutions from degradation. It is a better solution than wrapping a clear carboy in foil.
