The Caplugs® Evergreen is a 384-well flat-bottom black Polystyrene (PS) microplate — combining the 4× throughput advantage of 384-well format with the low-background fluorescence performance of black PS well walls. Black PS absorbs stray and reflected photons that create well-to-well cross-talk at the high well density of 384-well format — a more significant interference source than in 96-well format, where larger well spacing reduces optical cross-talk from adjacent wells. The delivers 5–20× lower fluorescence background versus natural or white 384-well plates for HTRF, TR-FRET, FRET, and fluorescence intensity assays at HTS scale.

Black 384-well is the industry standard format for HTRF (Homogeneous Time-Resolved FRET), LanthaScreen, and LANCE TR-FRET assays in pharmaceutical HTS — the low-background requirement for detecting the weak time-resolved donor fluorescence signal above plate autofluorescence is particularly critical at 384-well density where well cross-talk between adjacent high-signal wells is amplified. For absorbance ELISA, use natural 384-well (290-8217-010). 384-well edge-well evaporation is significant — use EZ-Seal™ film or lid for incubations >1 hour at 37°C.

Is the black 384-well the correct format for HTRF assays?

Yes. Black PS 384-well plates are the recommended format for HTRF (Cisbio), LanthaScreen (Thermo Fisher), and LANCE (PerkinElmer) TR-FRET assays. Black background eliminates donor fluorescence leakage and plate autofluorescence that interfere with the time-resolved emission window detection. HTRF assay developers universally specify black plates because the TR-FRET ratio (665 nm / 620 nm) is highly sensitive to background fluorescence at 620 nm — any plate material fluorescence in this window directly inflates the denominator and collapses the assay window.

What is the difference between black , white (290-8221-W10), and natural (290-8217-010) 384-well plates?

All three are 384-well flat-bottom PS plates in SBS standard footprint. Color determines the optical interaction with photons: Black absorbs stray light — minimizes fluorescence background and well cross-talk for FI, FRET, HTRF assays. Is the compatible with 384-well high-content screening (HCS)?

Yes. Black 384-well plates are widely used for high-content screening (HCS) and high-content imaging (HCI) where plate autofluorescence and well cross-talk must be minimized for sensitive immunofluorescence detection. However, note that standard PS 384-well plates are not TC-treated — for demanding adherent cell lines used in HCS, TC-treated variants provide better cell attachment consistency for imaging uniformity across all 384 wells.
