Technical Narrative

The Caplugs® Evergreen is a amber Polystyrene (PS) immunostain moisture chamber for IHC, IF, FISH, ISH, and other slide-based antibody incubation protocols requiring a controlled humid environment to prevent reagent evaporation during incubation. Evaporation of small-volume antibody solutions (10–100 µL) from slide surfaces during primary and secondary antibody incubation causes concentration gradients, uneven staining, edge artifacts, and false-positive signal — particularly in high-sensitivity assays such as phospho-antibody IHC, proximity ligation assays, and multiplex fluorescent panels. The moisture chamber eliminates these artifacts by maintaining a saturated humid environment around slides during incubation at room temperature, 4°C, or 37°C. Amber PS filters UV wavelengths, providing partial light protection for UV-sensitive antibody conjugates and fluorescent reagents without the complete light exclusion of the black variant — a practical choice for mixed-protocol laboratories handling both standard and light-sensitive IHC panels.

Virgin PS construction delivers autoclave compatibility for decontamination between staining runs, UV decontamination compatibility for clean-bench environments, and broad resistance to standard IHC reagents including PBS, TBS, citrate and EDTA antigen retrieval buffers, hematoxylin, eosin, and DAB chromogen. PS rigidity maintains chamber geometry under full slide loads without flex that could allow the lid seal to open during incubation and compromise humidity. The chamber floor is designed to hold distilled water or damp tissue at a defined depth below the slide surface, establishing consistent humidity without risk of fluid contact with the slide or tissue section.

At $142.48 per 3/Bag — $47.4905 per chamber — the delivers controlled-humidity incubation at a per-chamber cost that supports dedicated allocation per staining run or protocol. Also available: 240-9000-010 (PS Natural, $142.48/Bag); 240-9020-Z10 (PS Black, $142.48/Bag) — enabling matched procurement across all three light-handling specifications from a single vendor.

Immunostaining Workflow Compatibility —

The amber immunostain moisture chamber maintains the humid environment required across the following slide-based immunostaining and in situ hybridization workflows:

Application Specimen / Use Notes
Immunohistochemistry (IHC) Primary antibody incubation, overnight staining Humid environment prevents antibody solution evaporation from slides
Immunofluorescence (IF) Primary and secondary antibody incubation Prevents non-uniform staining from evaporation-driven concentration gradients
FISH / ISH Probe hybridization and washing steps Maintains humidity during extended hybridization incubations
H&E and special stains Extended incubation staining protocols Less critical than IHC but useful for high-humidity environments
CISH / chromogenic ISH Chromogenic probe incubation Same humidity requirements as FISH for even probe distribution
Proximity ligation assay (PLA) Antibody and ligation incubation PLA reagent volumes (10–50 µL) particularly vulnerable to evaporation
Phospho-antibody staining Phosphorylation-sensitive IHC panels Prevents evaporation artifacts that can mimic phospho-signal
Multiplex IHC / mIF panels Sequential antibody stripping and restaining Consistent humidity across all cycles reduces inter-cycle variability

Add distilled water or damp tissue to the chamber floor to establish humidity before placing slides. Do not flood the floor — capillary wicking onto the slide surface will dilute reagents.

Chemical & Environmental Resistance — Polystyrene (PS)

PS provides excellent resistance to aqueous IHC buffers and standard staining reagents. Avoid xylene and acetone — PS is solvent-sensitive.

Reagent / Condition Compatibility
Aqueous buffers (PBS, TBS, citrate) ✓ Excellent
Short-chain alcohols (EtOH, IPA 70%) ✓ Good — standard rinse concentration
Dilute acids (<10%) ✓ Good
Hematoxylin / eosin staining ✓ Compatible
DAB chromogen (brief contact) ✓ Good
Xylene ✗ Avoid — PS solvent sensitive
Acetone ✗ Avoid
Concentrated organic solvents ✗ Not recommended
Autoclave (121°C / 15 psi) ✓ Compatible
UV decontamination (254 nm) ✓ Compatible

Clinical & Research Laboratory Applications

The moisture chamber serves anatomic pathology and histology laboratories performing IHC panels for tumor diagnostics, biomarker profiling, and companion diagnostic testing; research laboratories running IF, FISH, and ISH protocols on tissue sections and cytology preparations; multiplex IHC and multispectral imaging programs requiring consistent antibody incubation across sequential staining cycles; proximity ligation assay workflows; phospho-protein IHC panels where evaporation-driven concentration errors are particularly impactful; and any pathology or research setting where slide-based antibody incubation is performed outside an automated stainer.

Frequently Asked Questions — Caplugs® Evergreen

Why does IHC require a moisture chamber — can't I just cover slides with a coverslip?

Coverslips prevent evaporation but create challenges with reagent distribution and antibody kinetics at low volumes. A moisture chamber allows antibody solutions to be applied directly to the slide surface (coverslip-free) at volumes of 50–200 µL, which is the standard manual IHC technique. The chamber maintains the humid environment that prevents evaporation of these micro-volumes during incubation — without the coverslip-related reagent trapping and distribution artifacts that complicate downstream washing steps.

What is the difference between the Natural, Black, and Amber moisture chambers?

All three are PS moisture chambers with identical dimensions and humidity performance. The difference is light handling: Natural (240-9000-010) is translucent — allows visual monitoring of slide moisture. Black (240-9020-Z10) completely excludes ambient light — use for photosensitive fluorescent secondary antibodies, TSA signal amplification, and fluorescent probe-based assays. Amber filters UV wavelengths for partial protection of UV-sensitive antibody conjugates in mixed-protocol labs. Choose based on the photosensitivity requirements of your reagents.

Can the moisture chamber be autoclaved between staining runs?

Yes. The is molded from Polystyrene (PS) and is autoclave-compatible at 121°C / 15 psi for decontamination between staining runs. PS is also UV decontamination-compatible for clean-bench environments. Note: PS is sensitive to xylene and acetone — do not use these solvents to clean the chamber.
