Why does IHC require a moisture chamber — can't I just cover slides with a coverslip?
Coverslips prevent evaporation but create challenges with reagent distribution and antibody kinetics at low volumes. A moisture chamber allows antibody solutions to be applied directly to the slide surface (coverslip-free) at volumes of 50–200 µL, which is the standard manual IHC technique. The chamber maintains the humid environment that prevents evaporation of these micro-volumes during incubation — without the coverslip-related reagent trapping and distribution artifacts that complicate downstream washing steps.
What is the difference between the Natural, Black, and Amber moisture chambers?
All three are PS moisture chambers with identical dimensions and humidity performance. The difference is light handling: Natural (240-9000-010) is translucent — allows visual monitoring of slide moisture. Black completely excludes ambient light — use for photosensitive fluorescent secondary antibodies, TSA signal amplification, and fluorescent probe-based assays. Can the moisture chamber be autoclaved between staining runs?
Yes. The is molded from Polystyrene (PS) and is autoclave-compatible at 121°C / 15 psi for decontamination between staining runs. PS is also UV decontamination-compatible for clean-bench environments. Note: PS is sensitive to xylene and acetone — do not use these solvents to clean the chamber.
